In de serie staat de Glee Club van de McKinley High School centraal. Deze club, waarin leerlingen dansen en zingen, stond ooit aan de top. Docent Will Schuester is vastbesloten de slapende Glee Club nieuw leven in te blazen en mee te doen aan de beroemde competitie The Nationals. Leerlingen en collega's verklaren hem voor gek, maar hij legt alle spottende reacties naast zich neer.

Na een oproep melden zich vijf leerlingen voor Glee Club. Het zijn allemaal outsiders, maar toch is Will enthousiast. Een zesde leerling weet hij met een valstrik binnen te halen. Ondanks de kritische houding van anderen doen de leden er alles aan om hun Glee Club tot een groot succes te maken.

Daarbij stuiten ze op de nodige weerstand van Sue Sylvester, coach van de cheerleaders. Zij moet een deel van haar budget met Will delen en is vastbesloten Glee Club ten gronde te richten.
